Amalie was born on 20 June 1754, at Prenzlau, in Brandenburg, Germany. She was the daughter of Henriette Caroline Christiane Louise of Pfalz-Zweibrücken (9 March 1721 - 30 March 1774) and Louis IX, Landgrave of Hesse-Darmstadt (15 December 1719 - 6 April 1790). Her maternal grandparents were Caroline of Nassau-Saarbrücken and Christian III, Duke of Zweibrücken. Her paternal grandparents were Charlotte of Hanau-Lichtenberg and Müntzenberg and Louis VIII, Landgrave of Hesse-Darmstadt. Amalie's parents were married on 12 August 1741, in Zweibrücken. Her siblings included: Caroline (1746-1821), Frederica (1751-1805), Louis X (1753-1820), Wilhelmina (25 June 1755 - 15 April 1776), Louisa (1757-1830), Frederick (1759-1802) and Christian (1763-1830). In 1773, when Catherine II the Great, Empress of Russia (1729-1796) was looking for a suitable wife for her son Paul (1754-1801), she sent an invitation to Amalie, Wilhelmina, Louisa, and their mother to visit Russia. Paul chose Wilhelmina to be his wife. She became Natalia Alexeievna and married Paul on 29 September 1773. Her mother, Henriette died on 30 March 1774. Amalie married Charles Louis, Hereditary Prince of Baden (14 February 1755 - 16 December 1801) on 15 July 1775, at Darmstadt, in Hessen. He was the son of Landgravine Karoline Luise of Hesse-Darmstadt (11 July 1723 - 8 April 1783) and Margrave Charles Frederick. Her sister, Natalia died on 15 April 1776, in St. Petersburg, Russia. Amalie and Charles Louis had eight children.

Her father, Louis died 6 April 1790. When Catherine, Empress of Russia was looking for a bride for Grand Duke Alexander Pavlovich she invited her daughters, Princess Louise and Princess Frederica to Russia. In the fall of 1792, the two sisters arrived in St. Petersburg. Louise learned Russian and converted from the Evangelical faith to Russian Orthodoxy on 9 May 1793, in St. Petersburg, receiving the name Elizabeth Alexeievna. Elizabeth married Grand Duke Alexander Pavlovich on 28 September 1793, at the Winter Palace, in St. Petersburg, Russia. Catherine the Great died in November 1796. She was succeeded by her son, as Paul I. Her daughter, Friederike Karoline Wilhelmine married Duke Maximilian of Zweibrucken on 9 March 1797. Her daughter, Frederica Dorothea Wilhelmina married Gustaf IV Adolf, King of Sweden on 31 October 1797 in Stockhom, Sweden. He was the son of Sophia Magdalena of Denmark and Gustaf III, King of Sweden. Elizabeth gave birth to a daughter, Grand Duchess Maria Alexandrovna on 29 May 1799. "As of this morning, I no longer have a child, she is dead," Elizabeth wrote to her mother on 27 July 1800. Her husband, Charles Louis died on 16 December 1801, in Arboga, Sweden. Her daughter, Marie Elisabeth married Friedrich Wilhelm, Duke of Brunswick-Wolfenbüttel on 1 November 1802. Elizabeth gave birth to a second daughter on 16 November 1806. The little girl died of an infection blamed on teething in 1808. Elizabeth wrote to her mother, "Now, I am not longer good for anything in this world, my soul has no more strength to recover from this last blow." Her son-in-law, Alexander I died of typhus on 1 December 1825. Elizabeth wrote to her mother on 4 December 1825, "Do not worry too much about me, but if I dared, I would like to follow the one who has been my very life." Elizabeth died on 16 May 1826. Amalie of Hesse-Darmstadt died aged 78, on 21 June 1832, in Bruchsal.
